Diisopropylamine dichloroacetate ( N-propan-2-ylpropan-2-amine ) is a kind of organic matter, the chemical formula is C8H17O2NCl2, white powder, bitter taste, soluble in water, ethanol or chloroform, slightly soluble in ether, almost insoluble in petroleum ether.
Diisopropylamine dichloroacetate is mainly used in patients with fatty liver in clinic. It has the function of liver protection and is an adjuvant drug for the liver. It can be used for cholestasis liver disease, acute and chronic hepatitis, fatty liver, early cirrhosis, jaundice and general liver and other causes of liver dysfunction. Diisopropylamine dichloroacetate ( DIPA ), alias Ganle, Liganneng, DADA, is an active ingredient of vitamin B15. The four methyl groups of DIPA can supply the body for the synthesis of choline. The main pharmacological action of DIPA is to regulate lipid metabolism disorders and provide methyl for choline and membrane phospholipids. The effect of dichloroisopropylamine acetate on fat is mainly by inhibiting the activity of citrate lyase, inhibiting the synthesis of acetyl coenzyme A, which is the raw material of fatty acid and cholesterol synthesis in cell fluid, and reducing the synthesis of fatty acid and cholesterol. Diisopropylamine dichloroacetate also inhibits the activity of hydroxylmethylglutaryl CoA reductase, a key enzyme in cholesterol synthesis, thereby inhibiting cholesterol formation ; diisopropylamine dichloroacetate also inhibits hormone-sensitive triglyceride lipase activity in adipose tissue, reduces fat mobilization, and reduces blood free fatty acids and glycerol levels ; diisopropylamine dichloroacetate increases glucose oxidation and promotes tricarboxylic acid cycle by activating pyruvate dehydrogenase ( PDH ) complex activity to provide energy for hepatocytes
Diisopropylamine dichloroacetate was synthesized by directly salting diisopropylamine with dichloroacetic acid as raw material in ether, so that the quality of diisopropylamine dichloroacetate synthesized by this method meets the medicinal standard, and the yield is 92.5 % ( based on dichloroacetic acid ). Compared with the literature process, this method simplifies the operation, is easy to produce, and has high yield and stable quality.
