Manufacturing Information

Chlorhexidine Diacetate is a disinfectant widely used in healthcare, especially for hand and surface disinfection. It has broad-spectrum antibacterial activity and can effectively kill a variety of microorganisms such as bacteria, viruses and fungi.

The synthetic route of product:

Concrete synthetic route is as follows:

1. Preparation of 5-chloro-2-methoxyphenylacetate

5-Chloro-2-methoxyphenylacetate is the precursor compound of product, and its preparation method is as follows:

(1)

Take benzaldehyde and 5-chloro-2-methoxyphenylacetic acid, add it to sodium hydroxide solution, and reflux at 110°C for 3 hours to obtain 5-chloro-2-(benzoyloxy)phenylacetic acid .

(2)

Condensing 5-chloro-2-(benzoyloxy)phenylacetic acid and formyl chloride to obtain 5-chloro-2-(benzoyloxy)phenylacetic acid methyl ester.

(3)

Hydrolyzing methyl 5-chloro-2-(benzoyloxy)phenylacetate to obtain 5-chloro-2-methoxyphenylacetate.

2. Preparation of nucleosides:

Nucleosides are intermediates synthesized from Chlorhexidine Diacetate. According to literature reports, nucleosides obtained by different methods are different. Here's one of those methods:

(1)

Take 5-chloro-2-methoxyphenylacetic acid ester and excess ammonium sulfate, add them to sulfuric acid solution, and react at 85°C for 30min to obtain 4-amino-5-chloro-2-methoxyphenylacetic acid.

(2)

Then take nitric acid and 4-amino-5-chloro-2-methoxyphenylacetic acid for nitration reaction at 25°C for 1h. The resulting product was reacted with copper acetate solution to give 4-chloro-N,N'-bis(2-hydroxyethyl)benzamide.

(3)

Finally, 4-chloro-N,N'-bis(2-hydroxyethyl)benzamide and ammonium chloride were reacted in ethanol to obtain nucleosides.

The reaction mechanism of product:

The synthesis of it involves multiple reactions, and each reaction has its specific reaction mechanism.

1. Nitrification reaction:

The nitration reaction is to nitrate 4-amino-5-chloro-2-methoxyphenylacetic acid, and the reaction mechanism is as follows:

(1)

Nitric acid reacts with ammonium sulfate to produce nitric acid and sulfuric acid.

(2)

Nicotinic acid reacts with 4-amino-5-chloro-2-methoxyphenylacetic acid, a nucleophilic substitution reaction occurs, and a nitro group replaces a methoxy group to generate 4-amino-5-chloro-2-nitrobenzene acetic acid.

(3)

In the presence of copper acetate, 4-amino-5-chloro-2-nitrophenylacetic acid reacts with sodium hydroxide to undergo aromatic tautomerization to generate 4-chloro-N,N'-bis( 2-Hydroxyethyl) benzamide.

It should be noted here that the entire nitrification reaction needs to be carried out under relatively protected conditions to avoid side reactions or danger.

2. Nucleoside generation reaction:

The formation reaction of nucleoside is as follows:

Add 4-chloro-N,N'-bis(2-hydroxyethyl)benzamide and ammonium chloride together to ethanol, a nucleophilic substitution reaction occurs, and the chloride ion replaces one of the dihydroxyl groups, This produces N,N'-bis(2-hydroxyethyl)4,5-dichlorobenzamide.

The obtained N,N'-bis(2-hydroxyethyl)4,5-dichlorobenzamide reacts with sodium bisulfite to generate nucleosides.
