Lecithin is commonly found in egg yolk, animal brain, soybean, corn, cottonseed and other resources. In practical applications, it is mainly extracted from soybean and egg yolk. Extraction of high purity lecithin has always been an important topic in phospholipid research. The oil residue of soybean processing soybean oil contains 40%~50% phospholipid, and the content of lecithin in soybean lecithin is about 16%~20%. The extraction and separation methods of soybean lecithin mainly include:

1) Organic solvent method

The traditional production of lecithin uses organic solvent for extraction. Its principle is to separate the components of mixed phospholipids according to the difference in solubility in solvent. The solvents used are generally C1~C4 low-carbon alcohol, n-hexane, petroleum ether, ether, ethyl acetate, etc. The key of solvent extraction method is to find a good solvent or solvent system, and the temperature, amount of solvent and solvent concentration must be controlled during extraction. Organic solvent can be divided into single solvent extraction method and mixed solvent extraction method.

2) Organic solvent inorganic salt composite precipitation method

This method is to extract lecithin from phospholipids by using the property that lecithin can precipitate with some inorganic salts, so as to separate it from other phospholipids and remove protein and fat. After comparing several inorganic salts, such as CdCl2, MgCl2, CaCl2, ZnCl2, used to extract soybean lecithin, it is considered that ZnCl2 is an ideal precipitator.

3) Chromatographic separation technique

Chromatographic separation is an efficient separation technology, which was mostly used in laboratories in the past, but has been gradually scaled up and applied to industry in the past 20 years. Thin layer chromatography, column chromatography and high performance liquid chromatography are commonly used in the extraction of soybean lecithin.

4) Thin layer chromatographic separation technology

Soybean lecithin was purified by centrifugal thin layer chromatography (CTLC) and identified by high performance liquid chromatography (HPLC) and thin layer chromatography (TLC). The results showed that 716mg pure SPC could be obtained with 1000mg injection volume. The CTLC method is simple and the preparation amount is large. The neutral impurities and other phospholipids are removed from the product, and the SPC purity is 99.99%.

5) Column chromatographic separation technology

Column chromatography uses the adsorbent as the stationary phase. When the solutes in the mobile phase pass through the stationary phase, their adsorption and desorption capacities are different, so as to achieve the purpose of separation. Li Wei et al. [8] used silica gel as adsorbent and mixed solution of methanol and chloroform with gradient difference of (1 ∶ 2)~(2 ∶ 1) to perform concave gradient elution, which can elute lecithin. The amount of eluant is only 5~6 times of the column volume, and the elution time is only 6h.

 

6) Supercritical extraction technology

Supercritical fluid extraction (SFE) is a new technology developed rapidly in recent years. The commonly used CO2 supercritical fluid can fully retain the nutritional and functional characteristics of the product. Through orthogonal experiment, the optimum technological conditions for preparing lecithin from Polygonum multiflorum Thunb. by supercritical CO2 extraction were studied. The optimum process conditions were selected as follows: extraction pressure 32MPa, extraction temperature 50 ℃, reactor pressure 6MPa, reactor temperature 55 ℃.

 

7) Membrane separation method

This method is to separate the lecithin from the mixture according to the molecular weight of different components in the lecithin and their difficulty in passing through the semi permeable membrane. If the phospholipid solution dissolved in the mixture of ethane and isopropanol passes through the polypropylene semi permeable membrane, the concentration of lecithin can be increased from 25% to 51%.
