On October 15, 2025, Eli Lilly and Company announced the positive top line results of two Phase 3 clinical trials, ACHIEVE-2 and ACHIEVE-5. ACHIEVE-2 is the second head to head controlled study in this research series, which evaluated the efficacy of orforglipron versus the SGLT-2 inhibitor dagelin in the treatment of type 2 diabetes adults with poor glycemic control by the two channels. ACHIEVE-5 evaluated the efficacy of orforglipron versus placebo in type 2 diabetes adults with poor glycemic control after titrated insulin glargine treatment (with or without DMD and/or SGLT-2 inhibitors).
At the 40th week of the two studies, under the goal of effectiveness estimation and the goal of treatment plan estimation, orforglipron (3mg, 12mg, 36mg) reached the primary and all key secondary endpoints, achieved significant A1C reduction, and reduced weight and improved a number of cardiovascular risk factors. The results of the two studies were consistent with the previously published research on type 2 diabetes.
