D-Salicin, also known as salicin, is a naturally occurring compound primarily found in the bark of white willow trees (Salix alba). It belongs to a class of compounds known as phenolics and more specifically, as salicylates. This compound has been of significant interest due to its structural similarity to aspirin, which is a synthetic derivative of salicylic acid.

Historically, it has been used in traditional medicine to alleviate pain, reduce inflammation, and treat fever. Its mode of action is believed to involve inhibiting the production of prostaglandins, which are hormones that contribute to inflammation and pain. However, unlike aspirin, it is not directly used in modern pharmacological preparations due to its slower absorption and metabolism.

Research has continued, exploring its potential benefits in various therapeutic areas, including as an anti-inflammatory agent, an analgesic (pain reliever), and even in the management of certain chronic conditions. Studies suggest that it may offer a natural alternative to synthetic drugs, with a comparably milder side effect profile.

Despite its promising properties, the use in medical treatments is still relatively limited and often requires further investigation to establish its efficacy and safety, especially in long-term use or in specific patient populations. Nevertheless, its traditional use and ongoing research highlight the potential of this natural compound in modern medicine.

Synthesis of Salicin via Esterification and Glycosidation
Step 1: Esterification of Salicylic Acid
  • Reaction Setup: Start with salicylic acid and an alcohol (commonly methanol or ethanol) in the presence of an acid catalyst, such as sulfuric acid or hydrochloric acid.
  • Reaction Conditions: Heat the mixture to a temperature ranging from reflux to around 100°C, depending on the alcohol used. Maintain this temperature for a few hours to allow the esterification reaction to proceed.
  • Isolation of the Ester: Once the reaction is complete, neutralize the acid catalyst using a base like sodium bicarbonate. Extract the ester from the reaction mixture using a suitable solvent. Concentrate and purify the ester by distillation or crystallization.
Step 2: Glycosidation with a Sugar Derivative
  • Reactant Preparation: Prepare the sugar derivative, typically glucose or galactose, in the form of an activated sugar ester or glycosyl halide. These activated intermediates are more reactive towards nucleophilic substitution reactions.
  • Reaction Setup: Combine the ester obtained from the esterification step with the activated sugar derivative in the presence of a catalyst, such as silver oxide or mercury(II) oxide.
  • Reaction Conditions: Heat the mixture to a temperature ranging from 60°C to 100°C for several hours to allow the glycosidation reaction to occur. The catalyst promotes the substitution of the ester group with the sugar moiety.
  • Isolation of Salicin: After the reaction is complete, quench the reaction mixture with water and extract the product with an organic solvent. Purify salicin by crystallization or column chromatography to obtain the final product.

Keratin regulation: bidirectional effect and precise penetration

1. Dissolve stratum corneum connections
By disrupting the desmosomes between keratinocytes, reducing intercellular adhesion, and promoting the shedding of aging keratinocytes. Its lipophilic properties allow it to penetrate the sebum layer, penetrate deep into the pores, dissolve accumulated keratin plugs (blackheads/whiteheads), and unblock hair follicle ducts. Experiments have shown that after treatment with 2% salicylic acid, the thickness of the stratum corneum is reduced by 15% -20%, and the renewal cycle of keratinocytes is shortened by 30%.

2. Bidirectional regulation mechanism
Low concentration (0.5% -2%): promotes keratinization metabolism, improves incomplete keratinization, suitable for keratosis (chicken skin) and ichthyosis.
Medium concentration (3% -5%): directly dissolves keratin, softens thick calluses and corns, clinically used for the treatment of palmar and plantar keratosis.
High concentration (10% -30%): As a chemical exfoliating agent, it stimulates epidermal reconstruction by disrupting the integrity of the stratum corneum, and is commonly used for acne scars and photoaging repair.

D-Salicin antibacterial | Shaanxi BLOOM Tech Co., Ltd

Anti inflammatory and antibacterial: multi-target inhibition of inflammatory cascade reaction

1. Inhibition of prostaglandin synthesis
By blocking the activity of cyclooxygenase (COX-1/COX-2), the production of prostaglandin E2 (PGE2) is reduced, thereby reducing vascular permeability and alleviating redness, swelling, and pain. Its selective inhibition of COX-2 (IC50=0.8 μ M) makes it significantly effective in the treatment of inflammatory acne, reducing the number of inflammatory papules by up to 40%.

2. Broad spectrum antibacterial effect
Propionibacterium acnes: By disrupting the structure of bacterial cell membranes and inhibiting their reproduction, the minimum inhibitory concentration (MIC) is 0.5% -1%.
Malassezia: With an inhibition rate of 85% against the pathogenic bacteria of seborrheic dermatitis, combined with its oil control effect, it can reduce the production of dandruff.
Resistance avoidance: Unlike antibiotics, salicylic acid kills bacteria through physical action and no bacterial resistance phenomenon has been observed.

Oil control and anti seborrhea: regulating sebaceous gland function

By inhibiting the activity of 5 α - reductase, the conversion of testosterone to dihydrotestosterone (DHT) is reduced, thereby lowering sebaceous gland secretion. Clinical trials showed that after 4 weeks of use of 2% salicylic acid gel, facial oil secretion decreased by 28%, and the effect lasted until 2 weeks after drug withdrawal. Its lipophilicity allows it to reside in hair follicles for a long time, continuously exerting its oil control effect.

Whitening and photoprotection: Multi pathway inhibition of pigment deposition

1. Inhibit tyrosinase activity
By competitively binding to the copper ion active center of tyrosinase, melanin synthesis is reduced. The inhibition rate is linearly related to the concentration, with an inhibition rate of 35% at a concentration of 2%.
2. Accelerate keratin metabolism
Promote the shedding of keratinocytes containing melanin and shorten the melanin transport cycle. Combined use of sunscreen can reduce UV induced pigmentation and increase the effectiveness of treating melasma by 25%.

faq

What is salicin used for?

Willow bark is commonly used to treat pain and reduce fevers. It contains a compound called salicin that has been shown to have anti-inflammatory effects.

Are salicin and salicylic acid the same?

In summary, salicin is usually milder and less irritating to the stomach lining because it requires metabolic conversion to exert its effects. Salicylic acid, on the other hand, is an active compound with stronger potency, often used topically for its immediate therapeutic properties.

What's another name for salicylic acid?

Salicylic acid is also known as 2-hydroxybenzoic acid (one of several β-hydroxy acids). It treats acne by causing skin cells to slough off more readily, preventing pores from clogging up as it is a keratolytic agent. Salicylic acid 1% to 2% preparations are usually well tolerated.

What is the difference between salicin and aspirin?
