Manufacturing Information-

Raw Material Pretreatment

 

 

High-quality walnut kernels are selected as raw materials, mechanically shelled, cleaned to remove impurities, and crushed to 40~60 mesh. Defatting is carried out via physical pressing or solvent extraction to obtain defatted walnut residue, which is then crushed to 45 mesh for later use.

Walnut Protein Extraction

 

 

Deionized water is added at a solid-liquid ratio of 1:8~1:12 (w/v). The pH value is adjusted to 8.5~9.0 with sodium bicarbonate or sodium hydroxide solution, followed by soaking at 55~65℃ for 2~4 hours. The mixture is centrifuged at 3000~5000×g for 15~20 minutes to remove precipitates, and the supernatant is collected. The pH is adjusted to 4.5 with citric acid or glacial acetic acid. After standing for 2.5 hours, centrifugation is performed again. The lower precipitate is washed with water until neutral to obtain walnut protein extract.

Compound Enzymatic Hydrolysis (Core Process)

 

 

A two-step dual-enzyme hydrolysis method is adopted: first, neutral protease (6000 u/g, accounting for 0.5%~2.0% of protein mass) is added for hydrolysis at pH 7.0 and 45℃ for 3 hours. After enzyme inactivation, alkaline protease (6000 u/g) is added for hydrolysis at pH 9.1 and 51℃ for 3.7 hours. Temperature, pH value and time are strictly controlled during enzymatic hydrolysis to ensure the hydrolysis degree exceeds 34%, yielding oligopeptides with molecular weight below 800 Da.

Filtration, Purification and Concentration

 

 

The enzymatic hydrolysate is filtered through an 80~100 mesh screen and centrifuged to remove impurities. Membrane separation technology (ultrafiltration/nanofiltration) is applied for purification to eliminate macromolecular proteins, polysaccharides and other impurities and enrich oligopeptides. Vacuum concentration is performed at 60~70℃ under -0.08~-0.1 MPa until the solid content reaches 20%~30%.

Drying and Finished Product Preparation

 

 

The concentrated liquid is processed by spray drying (inlet air temperature: 180~200℃, outlet air temperature: 80~90℃) or freeze drying to obtain off-white to light yellow powdered walnut oligopeptide. The product has a peptide content ≥70%, moisture ≤8%, and molecular weight mainly distributed in 200~800 Da.
