DeRuiZhi Pharmaceutical's oral small molecule GLP-1 has initiated Phase I clinical trials for the first time
On December 10, 2025, Dexmedetomidine registered a Phase III clinical trial of MDR-001 for obese or overweight subjects on the ClinicalTrials website. According to publicly available information, this is the first phase III initiated by the drug

This is a multicenter, randomized, double-blind, placebo-controlled phase III clinical study aimed at evaluating the efficacy and safety of oral small molecule drug MDR-001 tablets as an adjuvant therapy for lifestyle intervention in overweight or obese participants within 52 weeks. The objective of this clinical trial is to determine whether oral drug MDR-001 can improve weight management in overweight or obese adult participants. MDR-001 is a drug developed by DeRui Pharmaceutical with the help of Baiyan Molecular Pro" ™ An innovative drug pipeline designed with the assistance of a one-stop AI drug discovery platform.
The research results showed that the body weight of each dose group of oral MDR-001 tablets decreased by -8.2% to -10.3% compared to baseline, with an absolute weight loss of 7.4 to 9.2 kg, while the placebo group lost 2.4 kg. 70.9% to 85.4% of subjects lost more than 5% weight, and 34.5% to 48.1% of subjects lost more than 10% weight.
In terms of safety, MDR-001 tablets showed good safety and tolerability, with no serious adverse events (SAEs) related to drug treatment during the trial period. The main adverse events in each treatment group of MDR-001 were gastrointestinal reactions, including nausea, vomiting, and diarrhea, which were mostly mild to moderate and concentrated within 6 weeks before the titration period. The median recovery time was about 1-5 days, and the overall liver safety performance of the subjects was good. It is worth noting that the study included approximately 20% of subjects with a history of liver dysfunction and elevated transaminase levels in the treatment group. No trend of elevated transaminase levels was observed in any group, and compared to the placebo group, the transaminase levels in the MDR-001 treatment group were significantly reduced from baseline.
Zhengda Tianqing applies for listing of 'Simeglutide'
On December 11, 2025, CDE official website showed that Zhengda Tianqing's subsidiary, Lianyungang Runzhong Pharmaceutical Division, applied for the listing of megglutide injection, and its registered classification was 3.3 Zhengda Tianqing [Smegglutide], which applied for clinical treatment for the first time in June 2023, was approved for clinical treatment in August, and started clinical treatment for the first time in October.
At present, the drug has started two phase III clinical trials:
the indication for type 2 diabetes was started in January 2024, and the trial was completed in June this year;
The indication for obesity began in December 2024 and patient recruitment was completed in August of this year.
Simeglutide is a glucagon like peptide-1 receptor agonist (GLP-1 RA) initially approved for use in improving insulin resistance? The blood sugar control of type 2 diabetes patients was approved for the weight management of obese or overweight people due to the significant weight loss effect, and it was also proved to reduce the risk of major cardiovascular adverse events in type 2 diabetes patients. It simulates the natural action of GLP-1 in the human body, delaying gastric emptying, enhancing satiety to reduce food intake, and promoting insulin secretion and inhibiting glucagon secretion when blood sugar levels rise, achieving dual effects of lowering blood sugar and weight loss. The drug has a weekly injection type, with high compliance.
The common adverse reactions are mostly gastrointestinal related (such as nausea and diarrhea), and most of them are gradually tolerated with the time of administration. The original product of Smeglutide is developed by Nuo and Nuo De, and its approved indications include type 2 diabetes, cardiovascular risk, obesity and chronic kidney disease. In the first three quarters of this year, the global sales of the original product of Smeglutide have reached $25.5 billion.

Lilly Retarutide Phase III clinical data released, with a weight loss of 28.7% at 68 weeks
On December 11, 2025, Lilly announced that Retarutide, a phase III clinical trial TRIUMPH-4, achieved positive top line results. This trial aims to evaluate the safety and effectiveness of two highest study doses of retarutide for obese or overweight adults with knee osteoarthritis and no diabetes. The treatment method is to combine drugs on the basis of healthy diet and regular exercise. In this global registered clinical trial, 84.0% of participants had a baseline body mass index (BMI) of 235 kg/square meter. The results showed that both doses of retatrutide (9 mg and 12 mg) achieved all primary endpoints and key secondary endpoints. Based on two statistical methods of efficacy estimation and treatment plan estimation, after 68 weeks of treatment, the drug showed significant effects in weight loss, pain relief, and improvement of physical function. In terms of the common primary endpoint, retatrutide treatment achieved the highest average weight loss of 28.7% (approximately 32.3 kilograms, or 71.2 pounds): using the Western Ontario and McMaster University Osteoarthritis Index (WOMAC) pain score evaluation, the highest average pain level was reduced by 4.5 points, with a decrease of 75.8%

Retatrutide is a investigational triple hormone receptor agonist. As a single molecule drug, it can activate glucose dependent insulinotropic polypeptide (GIP), glucagon like peptide-1 (GLP-1), and glucagon receptor in the human body. Lilly is studying Retalutide in a number of phase III clinical trials to evaluate its potential efficacy and safety in obesity and overweight (with at least one weight related medical problem), type 2 diabetes, knee osteoarthritis, moderate to severe obstructive sleep apnea, chronic low back pain, cardiovascular and renal outcomes, and metabolic dysfunction related fatty liver disease.
