Pharmacological effects and antibacterial spectrum

(1) Pharmacology
Marbofloxacin tablets achieves bactericidal effect by inhibiting the activity of bacterial DNA gyrase (topoisomerase), thereby blocking the replication and transcription process of bacterial DNA. This mechanism of action enables mapofloxacin to exhibit strong antibacterial activity against various Gram positive and Gram negative bacteria.

(2) Antibacterial spectrum
The antibacterial spectrum of Mabofloxacin is extensive, including but not limited to the following strains:
Gram positive bacteria: Staphylococcus (including Staphylococcus aureus), Streptococcus, etc.
Gram negative bacteria: Escherichia coli, Salmonella typhimurium, Citrobacter freundii, Escherichia coli, Serratia marcescens, Morganella morganii, Proteobacter genus, Klebsiella genus, Shigella genus, Pasteurella genus, Haemophilus genus, Moraxella genus, Pseudomonas genus, Brucella canis, etc.
Mycoplasma: It also has good antibacterial effects against Mycoplasma.

In addition, mapofloxacin remains sensitive to some pathogens that have developed resistance to other antibiotics, such as Staphylococcus aureus, which gives it a unique advantage in treating complex infections.

Pharmacokinetic properties

Absorption and distribution

Mabofloxacin tablets are rapidly absorbed in dogs and cats, with a bioavailability close to 100%, indicating that the drug is almost completely absorbed in the gastrointestinal tract. Mabofloxacin has a low binding rate with plasma proteins (not exceeding 10%), which allows it to be widely distributed in the body and quickly enter various tissues and organs, including the liver, kidneys, skin, lungs, bladder, and digestive tract. In most organizations, the concentration of mapofloxacin is higher than that in plasma, which helps it exert stronger antibacterial effects at the site of infection.

Metabolism and excretion

Mabofloxacin is mainly metabolized by the liver in the body, converting into two inactive metabolites: N-demethylated MBF and N-oxo MBF. These metabolites and some prototype drugs are mainly excreted through the kidneys, of which about 2/3 are excreted through urine and 1/3 through feces. The elimination half-life of Mabofloxacin in dogs and cats is 14 hours and 10 hours, respectively, which allows its antibacterial effect to last longer and reduces the frequency of administration.

Pharmacokinetics in special populations and states

Pregnancy and lactation: In experiments on pregnant rats and rabbits, no side effects of mapofloxacin were found in pregnant animals. However, due to the lack of experimental research data on pregnant dogs and cats, this product should be used with caution during pregnancy. During lactation, after oral administration of mapofloxacin to sows, the elimination half-life of the drug is prolonged and the body clearance rate increases, which may require adjustment of the dosage.

Stability and Safety

Adverse reactions and contraindications

01/

Adverse reactions
When used at the recommended therapeutic dose, no serious adverse reactions were found in dogs and cats with Mabofloxacin tablets. Occasionally, mild adverse reactions may occur, such as vomiting, softening of stool, changes in drinking water, or temporary increase in activity, which usually disappear on their own after treatment. However, fluoroquinolone drugs may cause damage to the joint cartilage of puppies, so accurate dosage calculation and careful care should be taken when administering medication to puppies. In addition, fluoroquinolone drugs also have potential neurological side effects and should be used with caution in dogs and cats with epilepsy.

02/

Contraindications
Age restriction: Prohibited for dogs under 12 months of age or large dogs under 18 months of age (such as Great Danes, Borui dogs, Burnaby dogs, and Mastiffs), not recommended for cats under 16 weeks of age.
Special diseases: Animals allergic to fluoroquinolone drugs are prohibited.
Infection type: Not applicable to infections caused by anaerobic bacteria, yeast, or fungi.

Future prospects and research progress

Future prospects

With the increasing demand for antibiotics in veterinary clinical practice and the increasingly severe problem of drug-resistant bacteria, the application prospects of broad-spectrum antibiotics such as marbofloxacin tablets in veterinary clinical practice will be even broader. In the future, with the continuous advancement of drug development technology and the deepening of clinical applications, the antibacterial spectrum and pharmacokinetic properties of Marbofloxaci may be further optimized and improved to better meet the needs of veterinary clinical practice.

Research progress

In recent years, research on Marbofloxaci has been continuously deepening. On the one hand, researchers further validated its antibacterial activity and pharmacokinetic properties through in vitro and in vivo experiments; On the other hand, researchers are still exploring the combined effects of mapofloxacin and other drugs, as well as its potential in treating complex infections. In addition, with the increasingly prominent problem of drug-resistant bacteria, researchers are also paying attention to the antibacterial effect and resistance mechanism of mapofloxacin against drug-resistant bacteria.

Not recommended for use

Mabofloxacin is not omnipotent, and in some cases it may not be suitable for use. Below, I will explain in detail which situations it is not recommended to use Mabofloxacin. Everyone must pay attention!

1. Prohibited for small animals under 8 months old

Mabofloxacin is explicitly prohibited for small animals under 8 months old. Why? The main reason is that the cartilage and other important organ systems of these young animals have not fully developed, and their drug metabolism and excretion abilities are relatively weak. Mabofloxacin may have adverse effects on their cartilage and gastrointestinal tract, such as cartilage abnormalities, gastrointestinal reactions, etc. So, when using medication for young livestock, family members must be careful to avoid mapofloxacin!

2. Use with caution for animals with epilepsy

Although mapofloxacin is active against most gram-positive and gram-negative bacteria, including Pseudomonas aeruginosa, Klebsiella pneumoniae, etc., it also has weak effects on anaerobic bacteria and is mainly used for skin, soft tissue, and urinary tract infections. But its use in epilepsy treatment requires special caution. Mabofloxacin has a relatively long half-life, reaching 9-12 hours in dogs and up to 13 hours in cats. This means that the elimination of drugs in the body is relatively slow, which may increase the risk of drug accumulation in epileptic animals. Moreover, in the state of epilepsy, the excretory system of the affected animal may be affected, further affecting the normal metabolism and excretion of drugs. So, when using mapofloxacin to treat epilepsy in livestock, family members must be extra careful and closely monitor the occurrence of adverse reactions!

3. May lead to elevated liver enzymes

Liver enzymes are important indicators reflecting liver function, and their elevation often indicates damage to liver cells. The use of mapofloxacin may exacerbate this situation. In addition to drug factors, liver cell damage, inflammation, fatty liver, etc. may also lead to elevated liver enzymes. Mabofloxacin, as a drug, may directly damage liver cells, leading to an increase in liver enzymes. In addition, marbofloxacin tablets may also affect the metabolic function of the liver, leading to drug retention in the body and adverse reactions, further exacerbating liver damage. So, when using mapofloxacin, it is important to regularly monitor liver enzyme levels to ensure safe and rational use of the drug!
