The main component of tebufenozide powder is imidacloprid. The pure product is a white solid with the molecular formula C22H28N2O2 and CAS 112410-23-8. It has a melting point of 191 ℃, good solubility in water (20 ℃), and little solubility in other solvents. Manufacture Information

The preparation of tebufenozide powder requires two steps. First, the reagent n- (4-ethyl benzoyl) - n'- tert butyl hydrazine is synthesized, and then the reagent is used to synthesize tebufenozide.

Preparation of n- (4-ethyl benzoyl) - n'- tert butyl hydrazine:

 

Add 22.4 g of human tert butyl hydrazine hydrochloride (converted into hundreds), 50g of water, 120ml of solvent, 0.15g of self-made catalyst, control the reaction temperature to 0~5c, and protect it with nitrogen. Add 20% sodium hydroxide solution and 15% sodium carbonate solution successively for 0.5h. Then add solvent, and then add 28.8 g of 4-ethyl benzoyl chloride and 20% sodium hydroxide solution at the same time. The dropping time is 2 h, and then react for 1 h. Then raise the temperature to 60C, until all the solids are dissolved, and stand for layering. The organic layer was washed once with 80 g of 1% hydrochloric acid solution, stratified, and the solvent was removed to obtain 37.9 g of quasi white solid, with a content of 97.2% and a yield of 97.9%.

Preparation of tebufenozide:

 

Add the undissolved n- (4-ethyl benzoyl) - n'- tert butyl hydrazine solution prepared in 2.3.1 to a 500ml reaction flask, and control the kettle temperature at 60C. At the beginning, 3,5-dimethylbenzoyl chloride and base were added dropwise at the same time for 1 h, and then reacted for 3 h. Then add 50 g of water, raise the temperature to 92c until all the solids are dissolved, and let it stand for layering. The organic layer is washed once with 100g of 1% hydrochloric acid solution. The organic phase crystallizes at about 10C for 1h, and the white solid is filtered. The filter cake is the technical drug. Filtrate recycling. The average content of filter cake after drying is 96.3% by liquid chromatography.
